DESCRIPTION =========== Independently design, perform and analyze specialized major mouse experiments, including planning injection and drug treatment dosing schedule. Design and conduct the injection of growth factors, drugs, antibodies and tissue harvesting of mice. Perform orthotopic injection of tumor cell lines into various sites including spleen, liver, lung, mammary fat pad and underneath the kidney capsule. Design and conduct surgeries, experiments and euthanizing of small animals. Process tissues from mouse surgeries for cryopreservation and fixation. Perform mouse colony management such as breeding, weaning, monitoring and genotyping. Independently design, perform and analyze various cell biology techniques, cell culture techniques, and protein chemistry techniques. Design and conduct specialized gel electrophoresis techniques to evaluate protein modifications. Make significant innovative contributions pertaining to experimental design and methodology by reviewing published scientific data in refereed journals, as well as online using search engines and subject-specific websites, to make recommended changes to current protocols and practices. Develop novel protein purification and analysis techniques. Conduct sectioning of paraffin embedded and frozen tissues using microtome and cryotome equipment; perform histological and immunohistological staining techniques to evaluate protein localization in cells or tissues. Perform immunohistochemical analyses of frozen and fixed tissues or cells. Employ and develop novel imaging techniques to monitor tumor growth/metabolism. Troubleshoot problems for quality control on immunochemical and histochemical procedures. Maintain and prepare accurate records of all experiments. Assist in the preparation of data for publication. Provide training, work direction and supervision in areas of expertise as needed to all students and postdoctoral fellows. MINIMUM QUALIFICATIONS ====================== * Theoretical knowledge of molecular biology, immunology, cell biology, histology or related field as typically attained by a Bachelor's degree or an equivalent combination of education and experience. * Demonstrated strong experience and knowledge in coordinating major mouse experiments, such as planning injection and drug treatment dosing schedule. * Advanced knowledge in tissue culture techniques, including specialized assays such as cell migrations assays, cell-substrate adhesion assays, cell-cell adhesion assays immunocytochemistry and transfection. * Strong demonstrated experience using laboratory equipment such as centrifuges, spectrophotometers, balances, pH meters, autoclaves and electrophoresis equipment. * Strong demonstrated experience performing DNA and protein characterization techniques using electrophoretic analysis. * Demonstrated experience with protein biochemistry techniques; including quantification, ELISA assays, SDS PAGE, Western and Immunoblotting, phosphorylations and gel electrophoresis. * Demonstrated experience performing sterile eukaryotic cell culture techniques, PCR and agarose gel electrophoresis. * Demonstrated experience in sectioning of paraffin embedded and frozen tissues with microtome and crytome equipment. * Demonstrated experience using histological and immunohistochemical techniques to evaluate protein localization in fixed cells or tissues. * Demonstrated experience performing light, fluorescence and confocal microscopy techniques to visualize protein expression in tissue sections. * Demonstrated experience handling small mouse pups and retinal injection with Hamilton syringes. * Demonstrated experience in proper handling of chemical and biohazard materials. Demonstrated experience with sterile techniques. * Experience with computer software such as Excel to analyze data, store results and compute raw data. * Demonstrated experience of computer applications such as Word, Excel and PowerPoint. Internet search and software analysis capabilities. * Demonstrated experience handling and working with small animals, including demonstrated experience conducting mouse surgeries, experiments, and injections. * Knowledge in conducting euthanization of small animals. * Demonstrated experience ordering and maintaining lab equipment and supplies. * Experience in troubleshooting problems for quality control on every immunochemical and histochemical procedures.
